BEACON BIIB122 in LRRK2 carriers

Will tell us whether the drug is safe in LRRK2 carriers and changes cell markers. It cannot show whether it slows Parkinson's.

In short

After the LRRK2 inhibitor failed in broad early Parkinson's, Denali is testing it in people who carry a LRRK2 mutation; data expected first half of 2027.

The research record

This is the project's file for this landmark, as written and checked. Sources are listed at the end; anything not yet confirmed is marked "Not yet verified".

Who

Denali Therapeutics, which continues the study alone after Biogen and Denali stopped BIIB122 development in idiopathic Parkinson's.

What is being tested

BIIB122 (a LRRK2 kinase inhibitor, from landmark 7) against placebo for 12 weeks, then open-label, in people with a pathogenic LRRK2 variant. Phase 2a, 50 people (registry NCT06602193), started 24 October 2024; status active, not recruiting; primary completion estimated March 2027. It follows the missed LUMA trial in landmark 145.

What would count as success

It is a short biology study: safety, drug levels and biomarkers of lysosomal pathway engagement. No clinical-slowing endpoint, so it cannot prove benefit.

The drug's Phase 1 and 1b studies are in landmark 229.
