Landmark 185 · Trial in people
VTX3232 (NLRP3 inhibitor)
Lowered inflammation markers in about 10 people over 28 days, with no comparison group. It shows the drug acts on its target, not that it helps.
In short
A 28-day, open-label study of an oral NLRP3 inhibitor in about 10 people with early Parkinson's lowered inflammation markers in blood and spinal fluid with no drug-related adverse events; it is not evidence of benefit, and a controlled trial is only planned.

The research record
This is the project's file for this landmark, as written and checked. Sources are listed at the end; anything not yet confirmed is marked "Not yet verified".
Who
Ventyx Biosciences (the registered sponsor is its subsidiary Zomagen Biosciences). On 7 January 2026 Eli Lilly agreed to buy Ventyx for $14 a share, about $1.2 billion, with closing expected in the first half of 2026 (Ventyx SEC filing); I found no report that it closed. Sanofi had a right of first negotiation on VTX3232 that had not yet started.
What happened
The NLRP3 "inflammasome" is a protein complex that releases inflammatory signals such as IL-1β and IL-18. Blocking it protected dopamine neurons in mice (landmark 55). VTX3232 is a tablet designed to reach the brain.
- Design (NCT06556173). Phase 2a, one site, no placebo, single arm. Registry: 11 enrolled; company: 10 treated. Registry: started 8 August 2024; finished 4 April 2025 (Alzforum says September 2024). 40 mg once daily for 28 days. Primary measure: safety.
- Results (company release, 17 June 2025). No drug-related adverse events and no serious ones. Levels of the drug in blood and spinal fluid stayed above the level needed to block IL-1β for 24 hours. IL-1β in blood, IL-18 in blood and spinal fluid, IL-6 and hsCRP (a general inflammation marker) fell. Alzforum adds that sTREM2, neurofilament light and amyloid-beta in spinal fluid were unchanged and microglial PET showed no acute change. The company also reported better motor and non-motor scores, but no numbers appear in what I could read and there was no placebo. Full data were shown at the October 2025 International Congress of Parkinson's Disease and Movement Disorders (Alzforum).
- Next. Ventyx said it was planning a larger placebo-controlled Phase 2 (Alzforum, 16 Dec 2025). I found no registry entry for it.
Why it moved us toward a cure
It showed in people that an NLRP3 blocker can reach the brain's fluid and switch down the pathway landmark 55 pointed to. That is a step before asking whether it slows disease. The Cambridge DAPA-PD trial (landmark 186) tests a second NLRP3 drug with a placebo.
Outcome
Open. Target engagement is shown; clinical benefit is not tested.
