Landmark 291 · Trial in people

VENT-02 (NLRP3)

The company stopped it early after 29 people and gave no reason or results. We cannot say if it worked.

In short

Ventus ended a 28-day placebo-controlled study of its brain-penetrant NLRP3 inhibitor in Parkinson's after 29 people, giving "sponsor decision" as the only reason, and has not published results.

The research record

This is the project's file for this landmark, as written and checked. Sources are listed at the end; anything not yet confirmed is marked "Not yet verified".

Who

Ventus Therapeutics (the registered sponsor is Ventus Therapeutics U.S.).

What happened

  • Study (NCT06822517). Phase 1b (the registry tags it Phase 1/Phase 2; the company calls it Phase 2a). Randomised, double-blind, placebo-controlled; one dose twice a day for 28 days, then 7 days of follow-up. Planned about 30 people with mild to moderate Parkinson's; 29 enrolled. First patient dosed March 2025 (company release, 10 March 2025). Primary: safety. Secondary: drug levels and markers in blood and spinal fluid, and motor and quality-of-life data from wearables.
  • Stopped. Registry status is Terminated, 9 October 2025, reason "Sponsor decision". Alzforum (17 December 2025) says the reason is not given and that the company's site still listed the drug as Phase 2. The study had been meant to run to January 2026. Topline data had been guided for late 2025 or early 2026.
  • Earlier. In healthy adults (86 people, a Phase 1 that ran August 2023 to April 2024; company data via Alzforum) the drug reached the spinal fluid and fully blocked IL-1β release in blood samples.

Why it moved us toward a cure

It tests the same idea as landmark 55, a second brain-penetrant NLRP3 blocker after VTX3232 (landmark 185). Stopping does not show failure: with 29 people and 28 days it could say only about safety and target engagement. Roche's much larger study is landmark 290.

Outcome

Open. No results. I could not find why it stopped, so do not read it as a safety or efficacy failure.
